The Anti-ssDNA Antibody Test is a single-analyte blood test used to detect antibodies directed against single-stranded DNA (ssDNA). These antibodies are part of the broader family of anti-nuclear antibodies and can be found in a range of autoimmune conditions, including systemic lupus erythematosus (SLE), drug-induced lupus, and other connective tissue disorders. This test is typically requested when a clinician is investigating unexplained symptoms such as joint pain, skin rashes, fatigue, or recurrent fever, or when monitoring a known autoimmune condition. It is not a stand-alone diagnostic test: results are interpreted alongside your clinical history, physical examination, and other laboratory findings.

What is measured

  • Anti-ssDNA antibody: detects and quantifies antibodies that bind to single-stranded DNA. Unlike anti-dsDNA antibodies, which are highly specific for SLE, anti-ssDNA antibodies are more broadly associated with autoimmune activity and can be elevated in several conditions. Your clinician will use the result in context to help build a clearer picture of your immune status.

Who this test is for

  • People with symptoms that may suggest an autoimmune process, such as persistent joint pain, unexplained rashes, mouth ulcers, hair loss, or fatigue.
  • Individuals with a known or suspected connective tissue disease who need additional immune profiling.
  • Patients being monitored for disease activity or response to treatment under specialist care.

Why it matters

Autoimmune symptoms can be vague and overlap with many other conditions. Measuring anti-ssDNA antibodies gives your clinician an additional, focused piece of information that can help refine the differential diagnosis, guide further testing, and support timely referral to a rheumatologist if needed. Used responsibly alongside other clinical and laboratory findings, it contributes to a more precise and personalised approach to your care.

No special preparation required.

For blood tests, we recommend drinking plenty of water beforehand to ensure smooth sample collection. If you are taking any regular medication, continue to do so unless advised otherwise by your doctor.

Important Clinical Information:

This test is a screening tool to monitor and support your overall health. It does not replace your GP or consultant.

Always discuss your test results, especially any abnormal results, with your GP, who can interpret them in the context of your medical history and overall health.
