The Anti-dsDNA Antibodies Test measures antibodies directed against double-stranded DNA (dsDNA), the genetic material found in the nucleus of your cells. These autoantibodies are a hallmark of systemic lupus erythematosus (SLE) and are used to support diagnosis, monitor disease activity, and identify patients at higher risk of lupus nephritis (kidney involvement). The test is requested for people with suspected or confirmed lupus, those with unexplained joint pain, rashes, or fatigue, and for ongoing monitoring of known SLE. It may also be used alongside other autoantibody tests such as ANA and complement levels to build a fuller picture of immune activity.

The test is performed on a simple blood sample using an immunoassay method, typically ELISA or a similar validated technique, which detects and quantifies anti-dsDNA antibodies in the circulation.

Why it matters
Anti-dsDNA antibodies are highly specific for lupus and often rise before or during a flare, making them valuable for tracking disease activity over time. Persistently raised levels, particularly alongside low complement proteins, are associated with a greater risk of kidney inflammation. Regular monitoring can help your clinician adjust treatment promptly, reduce organ damage, and support clinical management.
